Paolo Nespoli is a "space engineer" by training with extensive experience in the space program. In October 2007, he launched the space shuttle Discovery on flight STS-120. The main objective of flight STS-120 was to carry the Harmony module and attach it to the International Space Station (ISS).

For the second flight, he launched in Soyuz TMA-20 from Baikonur Cosmodrome on 15 December 2010 as the ship's flight engineer. After the usual two-day flight, the Soyuz docked with the International Space Station on 17 December. He remained on the ISS for over five months as the onboard engineer of Expeditions 26 and 27.

In June 2015, he began preparations for another flight to the ISS, this time as a member of Expedition 52/53. He launched into space on July 28, 2017 in Soyuz MS-05, landed on the ISS after a six-hour flight, and began work on the station.

Paolo Nespoli retired from his active career as an ESA astronaut in 2019. He spent a total of 313 days, 2 hours and 36 minutes in space.
